GrB-Fc-HMEL is a preclinical-stage recombinant fusion protein developed by AbBC Therapies. It is part of the company's Targeted Granzyme B Immunotherapy (TGI) platform and consists of human Granzyme B fused to an Fc domain and a targeting moiety specific for Chondroitin Sulfate Proteoglycan 4 (CSPG4), also known as Melanoma-Associated Chondroitin Sulfate Proteoglycan (MCSP). The therapeutic mechanism involves the specific binding of the fusion protein to CSPG4-expressing tumor cells, followed by internalization and the delivery of Granzyme B into the cytosol. Once inside the cell, Granzyme B triggers the apoptotic pathway, leading to targeted cell death. This agent is being investigated for the treatment of various CSPG4-positive malignancies, including melanoma, triple-negative breast cancer (TNBC), lobular breast cancer, gliomas, and squamous cell carcinomas.
