GrB-Fc-IT4 is a fully human granzyme B-based fusion protein developed by AbBC Therapies for the treatment of multiple solid tumors. The therapeutic consists of the IT4 single-chain variable fragment (scFv) targeting the Fn14 receptor (fibroblast growth factor-inducible 14) fused to the serine protease granzyme B as the cytotoxic payload, connected through an immunoglobulin G heavy chain (Fc) domain. The drug works by delivering granzyme B specifically to Fn14-expressing tumor cells, where it induces apoptosis through rapid, irreversible activation of the caspase cascade and mitochondrial depolarization. Fn14 is upregulated in many solid tumors compared to healthy tissues, making it an attractive therapeutic target.
