GrB-Fc-SD1 is a preclinical-stage recombinant fusion protein therapeutic being developed by AbBC Therapies for the treatment of pancreatic and ovarian cancers. The drug is composed of a human single-domain antibody (SD1) that specifically targets Region III of the mesothelin glycoprotein, fused to a Granzyme B-Fc construct. This fusion protein is designed to deliver the cytotoxic serine protease Granzyme B directly into mesothelin-overexpressing tumor cells. Upon internalization, Granzyme B triggers apoptosis via both caspase-dependent and caspase-independent pathways. Preclinical studies have demonstrated nanomolar-range cytotoxicity and significant in vivo efficacy in tumor xenograft models.
