Overview

GRC-17536 is an orally available, potent, and selective small molecule inhibitor of the Transient Receptor Potential Ankyrin 1 (TRPA1) channel. It demonstrates high selectivity (over 1000-fold) for TRPA1 compared to other TRP channels, GPCRs, enzymes, and ion channels. The drug has shown efficacy in preclinical and clinical studies for neuropathic pain—particularly diabetic peripheral neuropathy—and as an anti-tussive agent in chronic cough conditions. It has also been investigated for inflammatory lung diseases such as allergic asthma. Developed originally by Glenmark Pharmaceuticals, it is considered a first-in-class selective antagonist of TRPA1[1][3][4][5][6][8].
