Overview

GRC 54276 is a small molecule inhibitor of Hematopoietic Progenitor Kinase 1 (HPK1), designed to enhance the immune system's ability to fight cancer by blocking HPK1-mediated negative regulation of T-cell activation. It is being developed as both monotherapy and in combination with immune checkpoint inhibitors, such as pembrolizumab or atezolizumab, for advanced solid tumors and lymphomas. Atezolizumab is a monoclonal antibody that targets programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 and CD80 receptors on T-cells, thereby promoting anti-tumor immune responses. The combination aims to synergistically boost anti-tumor immunity by inhibiting two distinct immunosuppressive pathways. GRC 54276 is under development by Glenmark, while atezolizumab (Tecentriq) was developed and is manufactured by Roche/Genentech[3][4][6][7].
