GRC 65327

GRC 65327 is a novel, selective, orally bioavailable small molecule inhibitor of Casitas B-lineage lymphoma b (Cbl-b), an E3 ubiquitin ligase that acts as a key intracellular regulator of T cell and natural killer (NK) cell activation. By inhibiting Cbl-b, GRC 65327 aims to enhance immune responses against tumors by reversing T-cell exhaustion and promoting robust immune cell activation in the tumor microenvironment. Preclinical studies have demonstrated its nanomolar potency, good in vitro and in vivo efficacy, and an acceptable non-clinical toxicity profile. The drug is being developed primarily for oncology indications—most notably triple-negative breast cancer—and has shown significant anti-tumor activity both as monotherapy and in combination with immune checkpoint inhibitors in preclinical models. It is currently under preclinical development through a collaboration between Ichnos Glenmark Innovation (an alliance between Ichnos Sciences Inc. and Glenmark Pharmaceuticals) and the National Cancer Institute[1][2][3][4].
