Overview

GREM1 siRNA (Gremlin-1 small interfering RNA) refers to small interfering RNA molecules designed to target and silence the expression of Gremlin-1 (GREM1). Gremlin-1 is a highly conserved glycoprotein and a member of the DAN family of bone morphogenetic protein (BMP) antagonists. It plays a critical role in embryogenesis, tissue differentiation, and organ formation, but its aberrant overexpression is implicated in various pathological conditions, including organ fibrosis (such as liver, pulmonary, and intestinal fibrosis), diabetic nephropathy, and several cancers (such as breast, lung, and colorectal cancers). By silencing GREM1, GREM1 siRNA restores BMP signaling (e.g., BMP-7 activity) and suppresses downstream profibrotic and pro-inflammatory pathways, such as TGF-β/Smad signaling. Preclinical research, including programs like University College Cork's nanoparticle-delivered GREM1 siRNA (DOP128), is exploring GREM1 siRNA as a therapeutic strategy for Crohn's disease-associated intestinal fibrosis, liver fibrosis, and oncology.
