GRI-0803

GRI-0803 is a novel, orally administered small molecule activator of human type 2 natural killer T (NKT) cells, also referred to as dNKT cells. It is being developed by GRI Bio for the treatment of autoimmune disorders, with an initial focus on systemic lupus erythematosus (SLE). The mechanism involves activation of type 2 NKT cells, which leads to dendritic cell-mediated inhibition of pro-inflammatory invariant NKT (iNKT) cells. This upstream immunomodulatory effect results in reduced inflammation and autoantibody production. In preclinical models, GRI-0803 demonstrated significant inhibition of lupus nephritis and improved overall survival without notable toxicology concerns or CYP450 interactions. The compound has favorable solubility and bioavailability profiles supporting once-daily oral dosing[1][4][6][9].
