Overview

GRIN2D siRNA is an experimental small interfering RNA (siRNA) designed to silence the expression of the *GRIN2D* gene, which encodes the GluN2D subunit of the N-methyl-D-aspartate (NMDA) receptor. Preclinical research indicates that GluN2D-containing NMDA receptors, particularly those located on GABAergic interneurons, play a critical role in the pathophysiology of depression and the mechanism of action of rapid-acting antidepressants like ketamine. By selectively knocking down the expression of the GluN2D subunit, GRIN2D siRNA inhibits NMDAR-mediated currents in interneurons, thereby modulating hippocampal inhibitory circuits and restoring the excitation-inhibition balance. In animal models, this targeted approach has been shown to produce rapid antidepressant-like effects and restore synaptic plasticity, suggesting that GluN2D inhibition may offer a therapeutic strategy for major depressive disorder with a potentially improved safety profile compared to non-selective NMDA antagonists.
