Overview

GRN-1201 + pembrolizumab is a combination therapy under investigation for the treatment of certain cancers, notably melanoma and non-small cell lung cancer (NSCLC). GRN-1201 is a cancer vaccine composed of four HLA-A2-restricted peptides derived from tumor-associated antigens (TAAs), designed to stimulate the immune system to recognize and attack tumor cells by activating cytotoxic T lymphocytes. Pembrolizumab is a monoclonal antibody that targets the programmed cell death protein 1 receptor (PD-1) on T cells, blocking its interaction with PD-L1 and thereby enhancing anti-tumor immune responses. The combination aims to synergistically boost anti-tumor immunity by both priming T-cell responses against specific tumor antigens (via GRN-1201) and preventing immune evasion by tumors (via PD-1 blockade with pembrolizumab). This regimen has been studied in clinical trials for metastatic PD-L1+ NSCLC and melanoma[1][2][3][4][5].
