Overview

**GRN-300 + paclitaxel** is a pharmaceutical combination regimen currently under investigation for recurrent or persistent, locally non-resectable or metastatic ovarian, primary peritoneal, and fallopian tube epithelial cancers. GRN-300 is a small molecule salt-inducible kinase (SIK) inhibitor administered orally, while paclitaxel is a well-established antineoplastic microtubule inhibitor given intravenously. The mechanism of GRN-300 involves inhibition of SIK2, which is implicated in the regulation of metabolic response and cell survival pathways; preclinical evidence suggests it may sensitize tumors to chemotherapy and other targeted agents. Paclitaxel stabilizes microtubules, inhibiting cell division. The combination is being tested in a phase 1/1B clinical trial for safety, pharmacokinetics, maximum tolerated dose, and preliminary clinical activity. GRN-300 is administered at the RP2D as determined in monotherapy dose escalation, combined with weekly paclitaxel (60 or 80 mg/m2). Exploratory translational objectives include biomarker analysis and response prediction[1][3][5][6][8].
