GRP78 siRNA is a small interfering RNA (siRNA) designed to silence the expression of **Glucose-Regulated Protein 78 (GRP78)**, also known as **HSPA5**. GRP78 is a molecular chaperone located in the endoplasmic reticulum that plays a pivotal role in the unfolded protein response (UPR) and cell survival under stress. In various cancers, including pancreatic ductal adenocarcinoma (PDAC), GRP78 is frequently overexpressed and can relocate to the cell surface, where it promotes tumor progression, chemoresistance, and activation of the PI3K/AKT signaling pathway. By utilizing RNA interference to knock down GRP78 levels, this therapeutic approach aims to inhibit pro-survival signaling and sensitize resistant tumor cells to chemotherapy agents such as gemcitabine.
