GRT-C901 is an investigational, patient-specific cancer vaccine developed as part of the GRANITE platform. It is an adenoviral vector-based priming vaccine that encodes tumor-specific neoantigens (TSNAs) identified from each patient's tumor using advanced computational prediction models. Upon intramuscular administration, the adenovirus infects cells and expresses these TSNAs, stimulating a cytotoxic T-lymphocyte (CTL) response against tumor cells presenting the same antigens. This immune activation aims to induce targeted lysis of cancer cells. GRT-C901 is typically administered in combination with a self-amplifying mRNA boost vector (GRT-R902) and immune checkpoint inhibitors such as ipilimumab and atezolizumab or nivolumab. The primary indication under investigation is metastatic microsatellite-stable colorectal cancer (MSS-CRC), but it has also been studied in other advanced solid tumors including non-small cell lung cancer, gastric cancer, bladder cancer, esophageal cancer, urogenital cancers, and general solid tumors[1][3][4][5][6][7].
