GRT-R912 + GRT-R914 + GRT-R918 is a combination of second-generation self-amplifying mRNA (samRNA) vaccine candidates developed by Gritstone bio for the prevention of COVID-19. These vaccines utilize a codon-optimized Spike cassette along with additional T cell epitopes (TCE) derived from highly conserved non-spike proteins (Nucleocapsid, Membrane, ORF3a, and Non-structural Proteins) to elicit broad and durable B cell and T cell immune responses. GRT-R912 and GRT-R914 target the Beta variant spike, while GRT-R918 targets the Omicron variant spike. The samRNA platform is designed to provide higher and more sustained protein expression at lower doses compared to conventional mRNA vaccines. The combination is currently being evaluated in the Phase 1 CORAL-CEPI clinical trial.
