GRT6015 is a small molecule inhibitor of the Tropomyosin receptor kinase A (TrkA), also known as NTRK1, developed by Grünenthal in collaboration with research partners such as Derphartox. It is primarily being investigated for the treatment of psoriasis, a chronic inflammatory skin condition. The Nerve Growth Factor (NGF)/TrkA signaling pathway is known to be upregulated in psoriatic lesions, where it contributes to keratinocyte hyperproliferation, increased nerve fiber density, and the recruitment of inflammatory cells. In preclinical studies using human lesional psoriasis skin explants, GRT6015 demonstrated the ability to modulate spontaneous cytokine expression, specifically reducing levels of pro-inflammatory mediators such as IL-17, IL-23, and TNF-alpha. This suggests that targeting the TrkA receptor may provide a novel therapeutic approach for managing the inflammatory cascade and neurogenic components of psoriasis.
