Overview

GRT7040 is a small molecule drug candidate developed by Grünenthal GmbH for the treatment of pain. It functions as a potent and selective agonist of the Nociceptin/Orphanin FQ (NOP) receptor, also known as the opioid receptor-like 1 (ORL1) receptor. While the NOP receptor is structurally related to classical opioid receptors (mu, delta, and kappa), it is functionally distinct. Agonism of the NOP receptor is believed to modulate nociceptive processing in the central nervous system, potentially offering analgesic efficacy with a improved safety profile compared to traditional mu-opioid agonists, particularly regarding respiratory depression and abuse liability. GRT7040 reached Phase 1 clinical trials to evaluate its safety, tolerability, and pharmacokinetics, but development appears to have been discontinued in favor of other candidates in the Grünenthal pipeline, such as the dual NOP/MOR agonist cebranopadol.
