GRTR775 is an investigational small molecule antagonist of the retinoic acid receptor-related orphan receptor gamma-t (RORγt), developed for the treatment of psoriasis. RORγt is a master transcription factor that regulates the differentiation of Th17 cells and the production of pro-inflammatory cytokines, including IL-17A, IL-17F, and IL-22. These cytokines are central to the inflammatory cascade in psoriasis, driving keratinocyte proliferation and skin inflammation. GRTR775 has been evaluated in preclinical studies, specifically using lesional psoriasis skin explant models in collaboration with Derphartox, where it demonstrated the ability to suppress spontaneous cytokine expression. By targeting the IL-17/IL-23 axis at the transcriptional level, GRTR775 represents a targeted approach to modulating the immune response in chronic inflammatory skin diseases.
