Overview

gruticibart (AB023, AB023-A2, xisomab 3G3) is a recombinant, humanized monoclonal antibody that targets the Apple 2 (A2) domain of coagulation factor XI (FXI). Developed by Aronora and Oregon Health & Science University, gruticibart selectively inhibits factor XIIa-mediated contact activation of FXI, while preserving the hemostatic feedback activation of FXI by thrombin. This unique mechanism of action allows the drug to prevent prothrombotic clot formation and device-initiated clotting (such as in hemodialysis circuits or central venous catheters) without impairing normal hemostasis or increasing bleeding risks. Gruticibart is currently in Phase 2 clinical development for the prevention of catheter-related thrombosis and for use in patients with end-stage renal disease undergoing hemodialysis.
