Overview

GS-0151 is an investigational therapeutic antibody developed by Gilead Sciences, originally discovered by MiroBio as MB-151. It is currently in Phase I clinical development for the treatment of rheumatoid arthritis. The drug acts as an agonist of Programmed Cell Death Protein 1 (PD-1), a checkpoint receptor expressed on activated T cells, B cells, and myeloid cells. By activating PD-1 signaling, GS-0151 downregulates immune cell activation—reducing T cell proliferation and pro-inflammatory cytokine production—and promotes apoptosis of autoreactive T cells. This mechanism aims to restore immune tolerance and mitigate autoimmune pathology in diseases such as rheumatoid arthritis. The approach represents a novel immunomodulatory strategy for autoimmune conditions[2][3][5].
