Overview

GS-0189 is a novel humanized IgG1 monoclonal antibody targeting signal regulatory protein alpha (SIRPα), a receptor for CD47 that delivers a "don't eat me" signal to macrophages. By blocking SIRPα, GS-0189 disrupts the CD47-SIRPα interaction and enhances phagocytosis of tumor cells by macrophages, resulting in direct antitumor effects. The drug was developed as an alternative to anti-CD47 therapies like magrolimab and was designed with an aglycosylated Fc region to prevent recognition by Fcγ receptors and avoid phagocytosis of SIRPα-expressing cells. It showed clinical activity in combination with rituximab in patients with relapsed/refractory non-Hodgkin lymphoma (NHL) but its development has been discontinued[1][3][6].
