GS-0201 + sacituzumab govitecan is an investigational combination therapy currently in Phase 1 trials for the treatment of advanced solid tumors. GS-0201 is a small molecule inhibitor targeting Poly (ADP-Ribose) polymerase 1 (PARP1), a key enzyme in DNA damage repair mechanisms, offering a targeted therapy approach for solid tumors[3][5]. This combination is designed to leverage dual mechanisms—PARP inhibition and targeted cytotoxic delivery—to improve antineoplastic efficacy in difficult-to-treat cancers[1][7]. GS-0201 is developed by Gilead Sciences and sacituzumab govitecan (Trodelvy) by Immunomedics, now a subsidiary of Gilead Sciences[2][5]. Primary indications include advanced malignant solid neoplasms, including metastatic breast cancer and other solid tumors[1][3][7].
