Overview

GS-073 is a small molecule inhibitor of microsomal prostaglandin E synthase-1 (mPGES-1) currently under development by Gesynta Pharma. The drug is designed to treat chronic inflammatory pain, specifically arthritis, by selectively reducing the production of prostaglandin E2 (PGE2). PGE2 is a key mediator of inflammation and pain, and its synthesis is catalyzed by mPGES-1 downstream of the cyclooxygenase (COX) enzymes. By targeting the terminal enzyme mPGES-1 rather than the upstream COX enzymes, GS-073 aims to provide potent anti-inflammatory and analgesic effects while sparing the production of other beneficial prostaglandins, such as prostacyclin. This selectivity is intended to reduce the risk of cardiovascular and gastrointestinal side effects commonly associated with traditional NSAIDs. GS-073 is currently in Phase 1 clinical trials.
