Overview

GS-1701 is an investigational, potent, long-acting small molecule inhibitor of the HIV-1 capsid protein, currently under development by Gilead Sciences. It belongs to a novel class of antiretrovirals that target the HIV-1 Gag polyprotein's capsid (p24) subunit. Unlike traditional antiretrovirals that target viral enzymes such as reverse transcriptase, integrase, or protease, GS-1701 disrupts multiple essential steps in the viral replication cycle. These include capsid-mediated nuclear import of the viral pre-integration complex, viral assembly and release, and the proper formation of the capsid core during maturation. Its development is part of a broader strategy to provide long-acting therapeutic and potentially prophylactic options for HIV-1, aiming for infrequent dosing intervals to improve patient adherence and quality of life.
