Overview

GS-221 is a small molecule inhibitor being developed by Grace Science for the treatment of various undisclosed cancer types. It specifically targets the NGLY1-NFE2L1 axis, a pathway that cancer cells utilize to survive proteasome stress. GS-221 works by inhibiting N-glycanase 1 (NGLY1), an enzyme essential for the processing and activation of the transcription factor NFE2L1 (also known as NRF1). NFE2L1 normally regulates the expression of proteasome subunits; when cancer cells are treated with chemotherapy or proteasome inhibitors, they often activate NFE2L1 to restore proteasome function. By blocking NGLY1, GS-221 prevents this 'bounce-back' response, effectively shutting down the cell's ability to regenerate its proteasome and thereby sensitizing the tumor to treatment.
