Overview

GS-4528 is an investigational monoclonal antibody developed by Gilead Sciences that targets interleukin-27 (IL-27). IL-27 is a heterodimeric cytokine belonging to the IL-12 family that plays a significant role in regulating immune responses, often exerting immunosuppressive effects within the tumor microenvironment. It acts by inducing the expression of multiple inhibitory receptors (such as PD-L1, LAG-3, and TIGIT) on T cells and promoting the differentiation of regulatory T cells (Tregs), thereby limiting effective anti-tumor immunity. By binding to and neutralizing IL-27, GS-4528 is designed to reverse these suppressive signals and restore the activity of cytotoxic T cells. It is currently being evaluated in Phase 1 clinical trials as a monotherapy and in combination with anti-PD-1 checkpoint inhibitors for the treatment of advanced solid tumors.
