Overview

GS-4774 is a heat-inactivated, yeast-based therapeutic vaccine designed to stimulate hepatitis B virus (HBV)-specific T-cell immune responses. It is engineered using recombinant Saccharomyces cerevisiae that expresses HBV X, surface, and core antigens. The primary goal of GS-4774 is to activate the host's immune system—specifically CD8+ T cells—to target and reduce the number of cells containing HBV in patients with chronic hepatitis B infection. Clinical studies have shown that while GS-4774 can enhance HBV-specific CD8+ T cell function and cytokine production (including IFNG, TNF, IL2), it did not produce significant reductions in serum HBsAg levels when used alone or in combination with standard antiviral therapy. The vaccine has been generally safe and well tolerated; injection site reactions are the most common adverse event[1][3][5][6][8][9].
