GS-4774 + tenofovir disoproxil fumarate is a **combination therapy** for chronic hepatitis B infection comprising **GS-4774**, a yeast-based therapeutic vaccine expressing HBV antigens designed to activate HBV-specific T cell immune responses, and **tenofovir disoproxil fumarate**, a nucleotide analogue antiviral agent that inhibits HBV replication. GS-4774 attempts to stimulate the host's immune system—particularly CD8+ T cells—by increasing production of cytokines such as IFNγ, TNF, and IL2 in response to HBV antigens. Tenofovir acts through direct inhibition of viral DNA polymerase as a small molecule antiviral. The goal of the combination is to both suppress HBV replication and enhance immune-mediated clearance of infected cells, potentially improving cure rates versus antiviral monotherapy.
