Overview

GS-5290 (formerly known as MBX-002) is an investigational, first-in-class monoclonal antibody agonist targeting the B- and T-lymphocyte attenuator (BTLA) receptor. Developed by Gilead Sciences following its acquisition of MiroBio in 2022, the drug is designed to restore immune homeostasis by selectively activating the BTLA inhibitory pathway. BTLA is an immune checkpoint receptor expressed on T and B cells that, when activated, provides inhibitory signals to dampen overactive immune responses. GS-5290 aims to mimic the natural inhibitory signaling of the BTLA pathway to treat autoimmune and inflammatory diseases. It is currently being evaluated in early-phase clinical trials for the treatment of ulcerative colitis, with the goal of inducing mucosal healing and clinical remission by suppressing pathological lymphocyte activation in the gut.
