GS-5737 is an inhaled small molecule inhibitor of the epithelial sodium channel (ENaC) developed by Gilead Sciences. It is being investigated for its potential to enhance mucociliary clearance (MCC) in patients with obstructive lung diseases, including cystic fibrosis and chronic obstructive pulmonary disease (COPD). By blocking ENaC-mediated sodium reabsorption in the airway epithelium, GS-5737 helps maintain airway surface liquid hydration, which is critical for effective mucus transport and lung defense. The drug is typically formulated in hypertonic saline to further support mucus hydration and clearance.
