Overview

GS-60667 (also known as GL60667) is a next-generation, long-acting HIV-1 capsid inhibitor developed by Gilead Sciences. As a small molecule, it targets the HIV-1 capsid protein (p24), which is essential for several critical stages of the viral replication cycle. GS-60667 works by binding to the capsid subunits and disrupting the stability and function of the viral capsid, thereby inhibiting the nuclear import of the viral pre-integration complex, as well as the assembly and maturation of new virions. This multi-stage mechanism of action provides potent antiviral activity and a high barrier to resistance. GS-60667 is being investigated for use in long-acting treatment and prevention (PrEP) regimens, potentially offering a more convenient dosing schedule compared to current daily oral therapies. It is currently in Phase 1 clinical trials to evaluate its safety, pharmacokinetics, and antiviral efficacy in humans.
