GS-9132, also known as ACH-806, is a first-in-class small molecule inhibitor of the hepatitis C virus (HCV) replication. Discovered by Achillion Pharmaceuticals and licensed to Gilead Sciences in 2004, the compound targets the HCV NS3 protease through a novel mechanism that differs from traditional active-site protease inhibitors. It was designed to inhibit viral RNA replication by interfering with the assembly or function of the HCV replication complex. While GS-9132 showed potent antiviral activity in genotype 1 HCV-infected patients during early clinical trials, its development was discontinued in 2006 following the observation of reversible elevations in serum creatinine in Phase I studies, suggesting potential renal toxicity.
