**GS-9721** is an investigational HIV broadly neutralizing antibody derived from **PGT121** and engineered to enhance Fc-mediated effector function and extend half-life. It binds the HIV-1 envelope, with activity directed against envelope components including **gp120** and functionally the **gp41-associated Env trimer complex**, and was developed by **Gilead** as part of HIV remission or cure-oriented combination strategies rather than as a marketed antiretroviral. Preclinical studies in SHIV-infected rhesus macaques evaluated GS-9721 together with the TLR7 agonist vesatolimod, where the Fc-enhanced antibody format was intended to improve antiviral immune clearance through stronger antibody-dependent cellular cytotoxicity and phagocytosis. Publicly available information indicates the program remained in early development and no approved commercial product or established clinical indication has been identified.
