Overview

GS-9911 is an orally bioavailable small molecule inhibitor of diacylglycerol kinase alpha (DGKα), developed by Gilead Sciences under license from Carna Biosciences. Its mechanism of action involves inhibiting DGKα, which enhances cytotoxic T cell activity and has potential immune checkpoint inhibitory and antineoplastic effects. By blocking DGKα, GS-9911 aims to potentiate T cell activation and promote immune-mediated tumor killing, making it a candidate for cancer immunotherapy. The drug is currently in Phase 1 clinical trials as monotherapy and in combination with anti-PD-1 monoclonal antibodies for the treatment of advanced solid tumors[1][2][3][5][7].
