GS24-B047 is a novel bispecific T-cell engager (TCE) designed to target Delta-like ligand 3 (DLL3) and the CD3 receptor complex on T cells, specifically for the treatment of small cell lung cancer (SCLC). It distinguishes itself from other DLL3-targeting agents, such as tarlatamab, by binding to a distinct, more membrane-proximal epitope on DLL3 and incorporating a proprietary integrated costimulatory signal. This costimulatory component is engineered to enhance T-cell activation, proliferation, and persistence while mitigating T-cell exhaustion in the tumor microenvironment. Preclinical studies have demonstrated that GS24-B047 possesses sub-nanomolar affinity for DLL3 and exhibits potent cytotoxicity against DLL3-expressing SCLC cell lines with a favorable therapeutic index. In vivo models have shown significant and durable tumor regression, supporting its further development in IND-enabling studies.
