GSC-DCV is an autologous dendritic cell (DC) vaccine in which patient-derived DCs are loaded with antigens from glioma stem-like cells (GSCs). The vaccine is designed to stimulate the immune system, particularly CD8+ T cells, to recognize and attack tumor cells in patients with recurrent glioblastoma multiforme (GBM). The process involves isolating DCs from the patient's blood, pulsing them ex vivo with lysates or antigens derived from GSCs—cells believed to drive tumor recurrence and resistance—and then re-administering these activated DCs intradermally. Clinical studies have shown that GSC-DCV can extend survival in recurrent GBM when used alone or in combination with anti-PD-1 monoclonal antibodies. The primary mechanism of action is enhanced antigen presentation by mature DCs leading to a targeted anti-tumor immune response[1][2][4].
