Overview

GSC-DCV + PD-1 blockade is a combination immunotherapy regimen under investigation for the treatment of recurrent glioblastoma (GBM). The therapy combines a dendritic cell vaccine (GSC-DCV), which uses patient-derived dendritic cells loaded with lysate from A2B5+ glioma stem-like cells, with a programmed cell death protein 1 (PD-1) immune checkpoint inhibitor. The GSC-DCV component aims to stimulate an anti-tumor immune response by presenting tumor-specific antigens to T cells, thereby activating cytotoxic CD8+ T lymphocytes. The PD-1 blockade component consists of monoclonal antibodies targeting the PD-1 receptor on T cells, preventing its interaction with PD-L1 and thus enhancing T-cell mediated anti-tumor activity by overcoming tumor-induced immunosuppression. This combination is being evaluated in clinical trials for safety and efficacy in patients with recurrent GBM who have previously undergone radiotherapy and chemotherapy[1][2].
