GSC-loaded autologous dendritic cells is an investigational cell-based cancer vaccine developed by the Fondazione I.R.C.C.S. Istituto Neurologico Carlo Besta for the treatment of recurrent glioblastoma. The therapy utilizes the patient's own dendritic cells, which are pulsed (loaded) with antigens derived from glioblastoma stem cells (GSCs) obtained from the patient's tumor tissue. By targeting GSCs, which are associated with tumor initiation, radio-resistance, and chemo-resistance, the vaccine aims to stimulate a robust immune response—specifically involving CD8+ T cells, NK cells, and NKT cells—to eliminate residual cancer cells and prevent recurrence. This approach is currently being evaluated in the Phase I DENDR-STEM clinical trial.
