Overview

GSH2 small interfering RNA is an experimental RNA interference (RNAi) therapeutic designed to silence the expression of the GS Homeobox 2 (GSH2, also known as GSX2) gene. GSH2 is a transcription factor that is frequently overexpressed in pancreatic ductal adenocarcinoma (PDAC) and is associated with tumor progression and resistance to chemotherapy, particularly gemcitabine. By utilizing the RNA interference pathway to degrade GSH2 mRNA, this agent reduces GSH2 protein levels. Preclinical studies in cell cultures and xenograft mouse models have demonstrated that silencing GSH2 inhibits cancer cell proliferation, migration, and invasion while promoting apoptosis. Furthermore, GSH2 silencing has been shown to alleviate gemcitabine resistance, a process mediated by the activation of the Sonic hedgehog (SHH)/GLI1 signaling pathway.
