Overview

GSI-136 is a small molecule γ-secretase inhibitor developed for the treatment of Alzheimer’s disease. It acts by inhibiting the γ-secretase complex, an intramembrane protease responsible for cleaving amyloid precursor protein (APP) and generating amyloid-beta peptides implicated in Alzheimer’s pathology. The drug was originally developed by Wyeth (now a subsidiary of Pfizer) and has been evaluated in phase I clinical trials to assess its safety, tolerability, pharmacokinetics, and pharmacodynamics in healthy subjects[1][2][3][4][5][6]. No evidence of further clinical development or approval beyond phase I has been reported.
