SC-24020, also known as GSK-3β Inhibitor I, is a potent, cell-permeable, and selective small molecule inhibitor of glycogen synthase kinase-3 beta (GSK-3β), with an IC50 of approximately 31 nM. It functions by inhibiting the kinase activity of GSK-3β, a key regulator in the Wnt/β-catenin signaling pathway and various other cellular processes including protein translation and apoptosis. While SC-24020 is primarily used as a research tool to study the biological roles of GSK-3β, it has been investigated in preclinical models of multiple myeloma and solid tumors to evaluate its potential to induce apoptosis or enhance the efficacy of proteasome inhibitors like bortezomib. Research has indicated that as a standalone agent, it may lack the potent apoptotic activity observed with dual GSK-3β/CDK inhibitors.
