GSK-3943104 is an investigational adjuvanted protein subunit therapeutic vaccine (targeted immunotherapy) developed by GSK for the treatment of recurrent genital herpes caused by herpes simplex virus type 2 (HSV-2) in already infected individuals.[3][7][13] The vaccine comprises recombinant HSV antigens formulated with proprietary adjuvants to elicit robust HSV-specific humoral and cellular immune responses, with the goal of reducing viral shedding, recurrence frequency, and severity of genital herpes episodes rather than preventing primary infection.[3][5][13] In the randomized Phase I/II TH HSV REC-003 first‑in‑human trial (NCT05298254), intramuscular administration of multiple non‑adjuvanted and adjuvanted formulations of GSK-3943104 demonstrated acceptable safety but failed to meet the primary efficacy endpoint in the Phase II part, leading GSK to terminate further clinical development and halt progression to Phase III, while continuing safety follow‑up.[1][5][8][11]
