GSK1247446A is an AS03-adjuvanted influenza vaccine candidate developed by GSK (GlaxoSmithKline). The vaccine utilizes the **AS03** (Adjuvant System 03) adjuvant, an oil-in-water emulsion composed of squalene, DL-alpha-tocopherol (vitamin E), and polysorbate 80, which is designed to enhance the magnitude, durability, and breadth of the immune response while allowing for antigen dose-sparing. GSK1247446A has been evaluated in two primary forms: a monovalent pandemic vaccine targeting the **H5N1** avian influenza strain (marketed as **Prepandrix** in the European Union) and a trivalent inactivated seasonal vaccine (**TIV-AS03**). Clinical trials, particularly in elderly populations, demonstrated that the AS03-adjuvanted formulation provided superior immunogenicity compared to standard non-adjuvanted vaccines like Fluarix. Although the H5N1 version received regulatory approval for pandemic preparedness, the seasonal trivalent version was not widely commercialized for routine use and development was eventually discontinued in favor of other seasonal vaccine platforms.
