Overview

GSK1940029 is a small molecule stearoyl-CoA desaturase 1 (SCD1) inhibitor developed by GSK as a topical gel for the treatment of acne vulgaris. SCD1 is an enzyme involved in the synthesis of monounsaturated fatty acids, which are essential for sebaceous gland function and implicated in acne pathogenesis. Preclinical studies demonstrated that topical application of GSK1940029 led to atrophy of sebaceous glands in mice, suggesting potential efficacy against acne. Phase I clinical trials evaluated its safety, tolerability, irritation potential, and pharmacokinetics after single and repeat topical dosing in healthy adults and patients with acne[1][3][4][7]. The drug was not found to be a primary or cumulative irritant at tested concentrations[1]. Development appears to have reached phase I with no recent reports of further advancement[7].
