GSK1995057 is a fully human, single domain antibody (domain antibody, dAb) developed by GSK as a potent and selective antagonist of the type 1 tumour necrosis factor receptor (TNFR1). It was designed to prevent pulmonary inflammation and treat acute lung conditions such as acute lung injury and adult respiratory distress syndrome. The drug acts by competitively inhibiting TNFR1, thereby blocking downstream pro-inflammatory signaling mediated by tumor necrosis factor alpha (TNF-α). GSK1995057 was evaluated in phase 1 clinical trials for safety, tolerability, pharmacokinetics, and pharmacodynamics via both inhaled and intravenous administration in healthy subjects.
