Overview

Recombinant WT1 Antigen-Specific Cancer Immunotherapeutic (GSK2130579A) is a cancer vaccine developed by GSK Biologicals in collaboration with the Institut Jules Bordet. It consists of a recombinant truncated Wilms' Tumor 1 (WT1) protein (WT1-A10) formulated with the AS01B adjuvant system, which contains the immunostimulants MPL (3-O-desacyl-4'-monophosphoryl lipid A) and QS-21 (a saponin fraction). The vaccine is designed to stimulate a specific immune response against WT1, a tumor-associated antigen that is highly overexpressed in acute myeloid leukemia (AML) and various other malignancies. Clinical investigation has focused on its use as a post-consolidation maintenance therapy in AML patients who have achieved complete remission, aiming to eliminate measurable residual disease and prevent relapse. Although Phase I/II trials demonstrated safety and the induction of humoral and CD4+ T-cell responses, the clinical development program was terminated by GSK due to changes in corporate strategy.
