GSK2189242A is an investigational pneumococcal protein subunit vaccine developed by GSK. Unlike traditional pneumococcal conjugate vaccines (PCVs) that target the polysaccharide capsule and are limited to specific serotypes, GSK2189242A is designed to provide serotype-independent protection by targeting three highly conserved surface proteins: Pneumococcal histidine triad protein D (PhtD), Pneumococcal choline-binding protein A (PcpA), and a detoxified version of pneumolysin (PlyD1). The vaccine was evaluated in clinical trials for the prevention of acute otitis media (AOM) and acute lower respiratory tract infections (ALRI) in infants, particularly in high-burden populations such as Native American communities. Despite completing Phase 2 evaluations, the program was ultimately discontinued by GSK in favor of other vaccine strategies.
