The pneumococcal protein candidate vaccine, identified by the development code **GSK2189242A**, is an investigational protein subunit vaccine developed by GSK. Unlike traditional pneumococcal conjugate vaccines (PCVs) that target the capsular polysaccharide, this vaccine is designed to provide serotype-independent protection by targeting highly conserved surface proteins of *Streptococcus pneumoniae*. The formulation contains two primary recombinant antigens: **detoxified pneumolysin (dPly)**, a modified version of the cholesterol-dependent cytolysin virulence factor, and **pneumococcal histidine triad protein D (PhtD)**, which is involved in metal ion homeostasis and bacterial adhesion. These antigens are adsorbed onto an aluminum-based adjuvant (alum). The vaccine has been evaluated in Phase I and Phase II clinical trials, including studies in infants and healthy young adults, to assess its safety, reactogenicity, and ability to induce broad immunity against invasive pneumococcal disease and pneumonia.
