GSK2189242A + GSK1024850A is an experimental combination pneumococcal vaccine developed by GSK Biologicals. It integrates the 10-valent pneumococcal conjugate vaccine (PCV10), marketed as **Synflorix** (GSK1024850A), with a protein-based vaccine component (**GSK2189242A**) containing two conserved pneumococcal proteins: detoxified pneumolysin (dPly) and pneumococcal histidine triad protein D (PhtD). Synflorix provides protection against 10 *Streptococcus pneumoniae* serotypes (1, 4, 5, 6B, 7F, 9V, 14, 18C, 19F, and 23F) by conjugating their capsular polysaccharides to carrier proteins, including Protein D from non-typeable *Haemophilus influenzae*. The addition of dPly and PhtD was intended to enhance the breadth of protection against non-vaccine serotypes by targeting virulence factors common across most pneumococcal strains. The combination was evaluated in Phase 2 clinical trials for the prevention of invasive pneumococcal disease and otitis media in infants and toddlers, but development was subsequently discontinued.
