Overview

GSK2193874 (often abbreviated as GSK219) is a potent and selective small molecule antagonist of the Transient Receptor Potential Vanilloid 4 (TRPV4) channel. Developed by GSK, the compound was primarily investigated for its ability to inhibit TRPV4-mediated calcium influx, which is a key regulator of vascular permeability and cellular responses to mechanical strain in the pulmonary system. Preclinical studies have demonstrated that GSK2193874 can reduce pulmonary edema and congestion in models of heart failure and acute lung injury. More recent research, including studies presented at ATS 2026, suggests that GSK219 can modulate neutrophil motility in the lung microenvironment, potentially mitigating the progression of emphysema and chronic obstructive pulmonary disease (COPD) by preventing neutrophil-mediated tissue damage in collapsed alveolar regions. While it reached early clinical development for heart failure-related pulmonary congestion, it has largely been utilized as a high-quality chemical probe in respiratory and cardiovascular research.
