GSK2197870A + pneumococcal conjugate vaccine

GSK2197870A + pneumococcal conjugate vaccine is an investigational combination vaccine candidate developed by GSK. It consists of a pneumococcal protein vaccine component (GSK2197870A), which contains two conserved recombinant proteins from *Streptococcus pneumoniae*—Pneumococcal histidine triad protein D (PhtD) and detoxified pneumolysin (dPly)—co-administered or formulated with a pneumococcal conjugate vaccine (typically the 10-valent PHiD-CV, brand name Synflorix). This dual-modality approach was designed to provide broader protection against pneumococcal disease than traditional conjugate vaccines alone. While the conjugate component induces serotype-specific antibodies against the polysaccharide capsule, the protein component (GSK2197870A) aims to induce serotype-independent immunity by targeting conserved virulence factors essential for colonization and invasive disease. Clinical development reached Phase 2 in infants and toddlers, but the specific combination was not progressed to Phase 3 as GSK shifted focus toward higher-valency conjugate vaccines.
